1. Weekly Fission Maze
2. Dailies then look for Nests on the Mission Board, run Nests with the rest afterwards.
3. Mission Board honestly. They seem to be pushing us to do them a lot now.
4. S Tier classes have been about the same for about the past year or so. Maybe a little variation here and there but mostly what they have been.

Can you be a little more specific for the last part? Because I am planning to create a sub chars

Honestly, just pick a class/classes you like and go with them.

If you don't have a good time with their play style then it doesn't matter how strong or good they are.

All characters in end game gear can be great.

But, as a personal recommendation, I'm partial to Moonlord, Saleana, Flurry and Saint.

But as said, any well geared character that you enjoy can be good.

Also, be sure to unlock your Mirage Jade.

Do mission board quests using multiple characters, 3-8 characters a day. (recommended to use same class with shared jades)
I earn more than 40k gold in 2 days by doing that. (unhealthy farming)

4. Top classes in this patch?
Same as last patch, I don't want to mention coz I might start a war if I said SB.

What gear are those characters using? Because I am starting to get the calypse first then run board quests so that I can at least run them on Lab 1 for the increased dungeon pts

You can get easy Calypse via Guide Mission by running each of the stated nest 5 times. Also, the increment of dungeon pts from increased lab is not that significant, where clearing the dungeon in lower lab for faster time is actually more efficient.
